In a preliminary trial with six stallions, the VetMotl Sperm Separation Device effectively enhanced post-thaw equine sperm quality by significantly improving mitochondrial activity, as measured by a double-stain fluorescence assay. This increase in mitochondrial activity sparked my interest for further investigation, as damaged mitochondria release pro-apoptotic factors that can harm DNA and lead to cell death, limiting sperm longevity after thawing and negatively affecting early embryonic development. Compared to traditional methods, VetMotl uniquely supported sperm vitality, despite similar motility. While no fertility data was collected, these findings suggest considerable potential for in vitro embryo production and broader field use in addressing stallion infertility, warranting further investigation to understand its full impact.

Since the first time a Brazilian distributor introduced a device that separates sperm using microfluidic technology, I was intrigued by the idea because of its effectiveness and simplicity. From then on, I began to think about how to explore its potential under field conditions, not just in labs for the well-known ICSI/IVF applications. The 3ml chamber opened several possibilities for improving stallion frozen/thawed samples and low-quality chilled doses as well, without the need for centrifugation. The results we’ve achieved in field practice and the data we’ve published support its feasibility, even for basic labs. We are currently exploring other applications to solve daily issues related to stallion semen in breeding routines, with promising outcomes to be published very soon.

Our laboratory has compared VetMotl microfluidic devices with traditional swim-up and centrifugation procedures for the isolation of fresh, cooled, and frozen-thawed stallion sperm for compatibility with assisted reproductive technologies such as in vitro fertilization. We find that these devices are undeniably superior for the selection of motile sperm and highly recommend these devices for sperm isolation procedures that require a selection of highly motile sperm.
